Can you file a lien for labor? Was hired to do work was not hired as a contractor.

Your question is broad, but generally laborers are allowed liens on property for doing work to improve it. Laborers are not required to serve a "notice to owner" the way subcontractors are, but laborers must file their liens within 90 days of finishing their work. There is no flexibility in this deadline.
How do I go about filing a lien for nonpayment of labor?

The construction lien form is found in section 713.08, Florida Statutes. I recommend that you consult with a lawyer to make sure that it is prepared correctly and to verify that you qualify for a laborer lien. The lien is filed with the clerk of courts in the county where the property is located. Here is a link to the statute: http://www.leg.state.fl.us/STATUTES/index.cfm?App_mode=Display_Statute&Search_String=&URL=0700-0799/0713/Sections/0713.08.html
